This previously unreported activity may enable novel approaches for characterization of miRNAs and mRNAs, including SNPs. SplintR is ideally suited for many target enrichment workflows with applications in next-generation sequencing and molecular diagnostics. The robust activity of the enzyme and its affinity for RNA-splinted DNA substrates (apparent Km = 1 nM) enable sub-nanomolar detection of unique RNA species within a complex mixture, making SplintR ligase a superior choice for demanding RNA detection technologies.<\/p>\n<p><strong>Applications<\/strong><\/p>\n<ul>\n<li>Ligation of ssDNA splinted by complementary RNA sequences<\/li>\n<li>Detection of RNA using ligation of DNA probes<\/li>\n<li>SNP or splice variant detection<\/li>\n<li>RASL-seq<\/li>\n<\/ul>\n<p>[\/vc_column_text][\/vc_column][\/vc_row][vc_row][vc_column][vc_single_image image=&#8221;1121&#8243; img_size=&#8221;full&#8221;][vc_column_text css=&#8221;.vc_custom_1432636896173{margin-top: -30px !important;}&#8221;]<span class=\"bildunterschrift\">(A) Outline of the ligation assay: a 5\u00b4-phosphorylated, 3\u00b4-FAM labeled DNA \u201cdonor\u201d oligonucleotide and an unmodified DNA \u201cacceptor\u201d oligonucleotide are\u00a0annealed to a complementary RNA splint. This substrate is reacted with a ligase to form a mixture of unreacted starting material (I), adenylylated DNA (II), and ligated product (III).
